Journey from deorphanisation to in vivo proof of concept
Drug development is based upon both Target Discovery and Drug Discovery; two challenging steps for researchers working toward GPCR-based drugs.  Our strategy for Target Discovery (e.g. GPCR deorphanization) will be exemplified to demonstrate the steps of tissue library preparation, screening readout, HPLC purification, surrogate hit identification and optimization, to the in vivo proof of concept. This process will be highlighted with specific receptors ( Free Fatty Acid Receptor 2 (GPR43) in the field of metabolic diseases and Epstein-Barr virus Induced GPCR 2 (EBI2) in the field of inflammation) and then also demonstrate various steps in Drug Discovery which provide in vitro pharmacology and SAR optimization for new target-specific NCEs.
